    Cheung 2 Lamberto Felida 2 Moniek P M de Maat 2 Ruud A Bank 3 Victor W M van Hinsbergh 1 and Pieter Koolwijk 1 1 Dept of Physiology VU University Medical Center Amsterdam 2 Dept of Hematology Erasmus Medical Center Rotterdam 3 TNO Quality of Life Gaubius Laboratory Leiden The Netherlands Tissue engineering is a new and upcoming field in which culture and application of living cells to replace or correct lost or damaged tissue is a great possibility for a number of health problems A challenge next to tissue engineering in vivo is in vitro culturing of cells in such a way that the cells stay undifferentiated or that they differentiate to a specific cell type Fibrin ogen forms a barrier to prevent blood loss next to that fibrin ogen can form a biological degradable matrix for the invasion and growth of cells Here we develop customized matrices containing variants of fibrin ogen for in vitro cell culture which is achieved by recombinant production of the fibrinogen variants A main goal of the study is to improve the efficacy of tissue engineering applications Expression vectors were constructed using mRNA of HepG2 cells and the corresponding cDNA sequences have been ligated in pcDNA3 1 vectors These vectors were used to transfect Chinese Hamster Ovary CHO cells with alpha beta and gamma chain of fibrinogen in order to obtain cells which produce intact recombinant fibrinogen Thus far we have obtained cells that produce the naturally occurring HMW and LMW fibrinogen variants Next to these naturally occurring variants fibrinogen with a decreased plasminogen binding and an increased resistance to plasmin degradation were designed The fibrinogen production was confirmed by ELISA and western blotting experiments The above described fibrinogen variants are produced by the CHO cells and will be tested on polymerisation
